HDAC6在ISO诱导大鼠心肌纤维化中的作用研究
Analysis of the Effect of HDAC6 on Myocardial Fibrosis Induced by ISO in Rats
摘要
Abstract
Objective The experiment was to investigate the expression of HDAC 6 in the rat model of myocardial fibrosis induced by ISO.Methods SD rats were randomly divided into 2 groups,each group of 20 which to set up the normal group and model group.The model group was treated with ISO daily by 5mg/kg for 7 d,the another group was given the same amount of saline.Take its blood and myocardial specimens.And tested its Heart quality index.ELISA was used to detected the content of type I and type III collagen.USed HE staining and Masson staining to observe the degree of myocardial fibrosis.The expression of HDAC6 andα-SMA were tested by Western blot.The mRNA expression ofα-SMA and HDAC6 were examined by qRT-PCR.Results Compared with model group,heart mass index was increased ( <0.05);HE staining and Masson staining showed collagen fiber hyperplasia in model group;Collagen type I and III in model group have increased relatively ( <0.05);Western blot showed that protein expression in model group was higher than normal group ( <0.05),α-SMA expression is obviously higher than normal control group ( <0.01);QRT-PCR detection showed that mRNA expression in model group was higher than normal group ( <0.05).Conclusion HDAC6 in plays an important role in development and incidence of myocardial fibrosis.关键词
