摘要

Abstract

Objective To explore the fuction of microRNA -34 c (miR-34c) in endometrial carcinoma.Methods The expression of miR-34 c in endometrial carcinoma and normal endometrial tissue was examed by reverse transcriptase polymerase chain reaction . RL95 -2 human endometrial carcinoma cells were divided into test group (miR-34c) and blank group (blank plasmid).Cell viability was meas-ured by MTT assay.Cell clone ability was assayed by clone formation assay.The cell cycle was measured by flow cytometry .The expression of cyclin-dependent kinase4 ( CDK4 ) and CDK6 , and the phosphorylation of retinoblastoma protein ( Rb ) were assayed by Western blot . Results The expression of miR -34 c in endometrial carcinoma tissues with 0.45 ±0.05 was lower than that in normal endometrial tissues with 1.03 ±0.10 ( P<0.01 ) .Compared with blank group , cell viability in test group was reduced with most reduced in fifth ( P<0.01 ) .The cell cycle was arrested at G1 phase was (57.75 ±5.45 )% in blank group and (78.73 ±8.62 )% in test group ( P <0.001 ).The expression of CDK4 and CDK6 was reduced ( P<0.01 ) .The phosphorylation of Rb was down regulated in test group ( P<0.01 ) .Conclusion miR-34 c expression could significantly inhibit the proliferation of endometrial cancer cell RL95-2.
