肿瘤微环境酸化下 MDSCs通过 ASICs 促进 TNF-α表达

摘要
Abstract
To investigate the relationship between the TNF-αexpression and ASICs in MDSCs under the tumor microenvironment , real-time PCR was performed to determine the TNF-αexpression changes in mice under normal physiological (pH 7.3), acidic (pH 5.5), and inhibitor-added condition, respectively.The results indicated that the expression of TNF-αsignificantly elevated in tumor MDSCs under normal physiological condition as well as acidic condition.In contrast, the expression was significantly inhibited by the addition of ASICs′nonspecific inhibitors, amiloride, diclofenac, or ASICs subunit 1 inhibitor PcTx-1.However, pretreatment with ibuprofen or ASIC subunits 3 inhibitors APETX2 had no effect .So the ASICs subunit 1 of MDSCs was likely responsible for promoting the expression of TNF-αunder the acidic tumor microenvironment .关键词
