摘要

Abstract

Objective:To study the miR-370 expression in hepatocellular carcinoma tissue and its relationship with clini-cal pathology and prognosis.Methods:Hepatocellular carcinoma tissue and normal tissue were collected to determine miR-370 and FOXO3a expression levels and analyze their correlation with clinical pathological characteristics;hepatocellular carcinoma cells were cultured and transfected with miR-370 mimics and FOXO3a-overexpression plasmids,and then cell proliferation and migration were detected.Results:miR-370 expression level in hepatocellular carcinoma tissue was significantly higher than that in normal tissue,FOXO3a expression level was significantly lower than that in normal tissue and miR-370 expression level was negatively correlated with FOXO3a expression level (P <0.05);miR-370 expression levels in hepatocellular carcinoma tissue&nbsp;with TNM Ⅲ~Ⅳ stage and vascular invasion were significantly higher than those in hepatocellular carcinoma tissue with TNMⅠ~Ⅱ stage and without vascular invasion,and FOXO3a expression levels were significantly lower than those in hepatocellular carcinoma tissue with TNM Ⅰ~Ⅱ stage and without vascular invasion (P <0.05);transfection of miR-370 mimics could pro-mote hepatocellular carcinoma cell proliferation and migration and reduce FOXO3a mRNA content;after transfection of miR-370 mimics and FOXO3a-overexpression plasmids,the effect of miR-370 mimics on promoting hepatocellular carcinoma cell proliferation and migration was weakened.Conclusions:miR-370 expression in hepatocellular carcinoma tissue significantly in-creases and is closely related to the TNM stage and vascular invasion of the tumor,and miR-370 can targeted inhibit FOXO3a expression so as to inhibit hepatocellular carcinoma cell proliferation and migration.
