Expression of Osteopontin and Matrix Metalloproteinase 9 in the Endometrium of Patients With Endometriosis and Influence of Progesterone on Their Expression

Abstract
Objective To investigate the expression and correlation of osteopontin ( OPN ) and matrix metalloproteinase 9(MMP - 9)in the endometrium of patients with endometriosis(EMs),and the influence of different doses of progesterone on their expression. Methods 105 patients with EMs,who received treatment in General Hospital of Ningxia Medical University from 2006 to 2015 and met the inclusion criteria,were selected in the study. The eutopic endometrium was got 1 d before surgery by curettage(EMs eutopic endometrium group). The ectopic endometrium was got through oophorocystectomy or tissue adnexectomy(EMs ectopic endometrium group). In addition,90 patients who had underwent sterilization operation of fallopian tube and the endometrium was got through curettage 1 d before sterilization operation( normal endometrium group). Immunohistochemistry was used to detect the expression status of OPN and MMP - 9 in the endometrium among the three groups, and the correlation of OPN and MMP - 9 between EMs eutopic endometrium group and EMs ectopic endometrium group were also analyzed. The eutopic endometrium of 9 patients were randomly selected from the 105 patients to carry out follow - up experiment, inverse transcription polymerase chain reaction(RT - PCR)was applied to detect the expression levels of OPN and MMP - 9 in the endometrial epithelial cells after progesterone intervention at different doses(0,1,10,100,1 000 nmol/ L). Results Immunohistochemistry results showed that the color of endometrium epithelial cells in eutopic endometrium after DAB staining in normal endometrium group was light yellow,and OPN and MMP - 9 were mainly expressed in endometrium epithelial cells. The staining of endometrium epithelial cells in EMs eutopic endometrium group darkened and presented brown. The staining of endometrium epithelial cells in EMs ectopic endometrium group darkened further and showed brownish black,and OPN and MMP - 9 were mainly expressed in EMs epithelial cells. The expression levels of OPN and MMP - 9 in EMs ectopic endometrium group were stronger than those in EMs eutopic endometrium group and normal endometrium group( u = 7. 553, - 6. 153,- 9. 754, - 10. 240,P ﹤ 0. 001);the expression levels of OPN and MMP - 9 in EMs eutopic endometrium group was stronger than those in normal endometrium group( u = 7. 164,8. 816,P ﹤ 0. 001). The result of Spearman correlation analysis was shown as follows:the expression levels of OPN and MMP - 9 between eutopic and ectopic endometrium group were positively correlated with each other(rs = 0. 657,0. 745,P ﹤ 0. 05). After intervention of progesterone at different doses of 0 nmol/ L,1 nmol/ L,10 nmol/ L,100 nmol/ L,and 1 000 nmol/ L,the expression levels of OPN and MMP - 9 were not significantly different(P ﹥ 0. 05). Conclusion Enhanced expression of OPN and MMP - 9 in eutopic and ectopic endmetrium of patients with EMs was presented,but different doses of progesterone has no significant influence on the expression level of OPN and MMP - 9 in the eutopic endometrium epithelial cells of EMs patients.关键词
