1型糖尿病大鼠模型的建立及相关病理学形态观察
Establishment of type 1 diabetic rat model and observation of related pathological morphology

Abstract
Objective To study the construction method of Streptozotocin(STZ)-induced type 1 diabetes mellitus(T1DM) rat model and to observe the related organs pathological morphology. Methods Forty-five cases of T1DM rat model were estab-lished by once intraperitoneal injection of different doses(50,70,90 mg/kg) of STZ,which were respectively served as the 50, 70,90 mg/kg groups,15 cases in each group;other 15 rats without injection of STZ were selected as the control group. The fasting blood glucose values of these rats were detected before the model construction and on 3 ,7,14,21 d after the model construction. All survival rats were killed on 21 d. The pancreas,heart,lungs and kidneys were taken for preparing the paraffin sections and per-formed the HE staining for conducting the optical microscopic observation. Results The rat blood glucose values of different doses of group reached the model standard. The 70 mg/kg group was survived on 21 d ,while the 50,90 mg/kg groups began to die on 7 d and the 21 d survival rates were 60.0%(9/15) and 13.3%(2/15) respectively;the pathological morphological changes of the rat pan-creas and kidneys in different doses of groups were obvious compared with those in the control group ,while which of heart and lung had no significant morphological changes. Conclusion Once intraperitoneal injection of 70 mg/kg for inducing DM rat model is more stable and the morphological changes of the pancreas and kidney are obvious.关键词
