Abstract

Objective To establish the heart failure model of rabbits by increasing the cardiac preload and afterload and to examine the role of DKK3 in the heart failure by detecting the expres⁃sion change of DKK3. Methods Twenty Japanese white rabbits were randomly divided into the con⁃trol group and the heart failure group. The rabbit aortic valve was punctured to cause reflux and the abdominal aorta was narrowed 2 weeks later in the heart failure group. Animals in the control group only underwent sham operation. After two months, the structure and function of the heart were meas⁃ured, the myocardial cell size and collagen reconstruction were evaluated by hematoxylin and eosin (HE) staining and picrosirius red (PSR) staining, and the changes of DKK3 were detected by immunofluorescence. Results The ejection fraction was significantly decreased, the heart weight, cell size and collagens were markedly increased and the expression of DKK3 was obviously decreased in the heart failure group compared with the control group. Conclusion The DKK3 expression is decreased in heart failure, which weakens the protective effect of DKK3 on cardiac remodeling.
