外源添加代谢中间产物对米根霉生产富马酸的影响
Effects of supplemented metabolic intermediates on production of fumaric acid by Rhizopus oryzae

Abstract
Effects of supplemented metabolic intermediates on production of fumaric acid and key enzymes were studied for Rhizopus oryzae fumaric-malic acid producing strain. R. oryzae could produce 34. 0 g/L fumaric acid when 4. 0 g/L malic acid was added in the medium,which was 24. 5% higher than that of the control. Addition of malic acid increased specific activity of fumarase( FUMR) and decreased specific activity of alcohol dehydrogenase(ADH)in R. oryzae. Adding less than 1. 0 g/L oxaloacetate increased specific activity of FUMR and improved fumaric acid production by R. oryzae. However,adding more than 1. 0 g/L oxaloacetate in the medium decreased both FUMR and ADH specific activities in R. oryzae, which resulted in a sharp decrease of fumaric acid and ethanol. The addition of 1. 5 g/L pyruvate improved fumaric acid production from 27. 3 g/L to 38. 7 g/L. Compared to the control,41. 8% increase of fumaric acid was observed. At the same time, a 34. 7% decrease of FUMR specific activity was observed with no change to ADH specific activity.关键词
