摘要

Abstract

Effects of supplemented metabolic intermediates on production of fumaric acid and key enzymes were studied for Rhizopus oryzae fumaric-malic acid producing strain. R. oryzae could produce 34. 0 g/L fumaric acid when 4. 0 g/L malic acid was added in the medium,which was 24. 5% higher than that of the control. Addition of malic acid increased specific activity of fumarase( FUMR) and decreased specific activity of alcohol dehydrogenase(ADH)in R. oryzae. Adding less than 1. 0 g/L oxaloacetate increased specific activity of FUMR and improved fumaric acid production by R. oryzae. However,adding more than 1. 0 g/L oxaloacetate in the medium decreased both FUMR and ADH specific activities in R. oryzae, which resulted in a sharp decrease of fumaric acid and ethanol. The addition of 1. 5 g/L pyruvate improved fumaric acid production from 27. 3 g/L to 38. 7 g/L. Compared to the control,41. 8% increase of fumaric acid was observed. At the same time, a 34. 7% decrease of FUMR specific activity was observed with no change to ADH specific activity.
