曼陀罗子对小鼠L5178Y细胞TK基因的致突变作用
Semen Daturae Induced TK Gene Mutation in Mouse L5178Y Cells
摘要
Abstract
Objective To investigate the TK gene mutation in mouse lymphoma cell induced by semen daturae. Methods L5178Y cell was treated for 3 h by 62. 5, 125, 250, 500 μg/mL of total alkaloids of semen daturae or 625, 1 250, 2 500, 5 000 μg/mL of scopo-lamine hydrobromide in the activation of metabolic and non-metabolic activation conditions. The supernatant fluid was discarded, and the DMEM media was added to continue developing for 48 h. The number of mutations cell colony in each group was calculated after L5178Y cell was cultured in 96 well plate with TFT. Results Compared with the control group, the mutation rates of L5178Y had no significant increased after total alkaloids of semen daturae or scopolamine hydrobromide treated. Conclusion 62. 5-500 μg/mL semen daturae and 625-5 000 μg/mL scopolamine hydrobromide have no mutation effect on TK gene in mouse lymphoma cell.关键词
