摘要

Abstract

Objective To detect the cell growth state and evaluate the biological indicators of senescence degree of human embryo lung diploid cells by growth curve and flow cytometry assay. Methods The Walvax-2 cells were splited into 16 generations to detect the cell proliferative ability and cell vitality in different generations of Walvax-2 cells by drawing growth curve.The cell cycle phase and cell apoptosis were analyzed by fluorescence activating cell sorter (FACS)assay on different generations Walvax-2 cells with propidium iodide(PI). Results The cell vitality of P20 and P30 were stronger than the P40 and P50 (all P <0.05),and that of P40 was stronger than the P50 (P >0.05).The cell percentage of G0 /G1 phase has no association with the generations of Walvax-2 cells (P >0.05),however the synthesis phase,G2 /M phase and cellular debris were significantly associated with the generations of Walvax-2 cells growth (all P <0.05). Conclusion Drawing growth curve can distinguish cell vitality of Walvax-2 cells which were separated by large generations.The cell cycle changes are detected by fluorescence activating cell sorter assay with propidium iodide and it can act as a biological indicator to evaluate the senescence degree of Walvax-2 cells.
