Production of feruloyl esterase by solid-state fermentation of Aspergillus niger and preparation of ferulic acid from wheat bran by enzymatic hydrolysis

阿魏酸是一种广泛存在于植物细胞壁中的羟基肉桂酸,在医药、食品等行业具有巨大的应用前景。笔者选取黑曲霉菌株TRIIM 3�00954固态发酵产生阿魏酸酯酶,并研究了其酶学性质和酶解麸皮制备阿魏酸的能力。结果表明,以麦麸和甘蔗渣质量比2∶1混合作为基质,添加0�6%葡萄糖作为补充碳源,1�0%尿素为补充氮源,初始pH 6�0,30℃发酵培养7 d后,获得的阿魏酸酯酶产量最高为8�2 U/g。粗酶液中阿魏酸酯酶最适温度和pH分别为50℃和5�0。以0�6 U/g的阿魏酸酯酶和90 U/g的木聚糖酶联合作用去淀粉麸皮,50℃和pH 6�5下酶解6 h,最终可将去淀粉麦麸中60�6%的阿魏酸释放出来。利用农林加工副产物提取酚酸类物质、提升产品附加值是近年研究热点,本研究建立的阿魏酸酯酶发酵及酶法制备麦麸阿魏酸的技术,为工业化提取阿魏酸提供了有效途径。

Ferulic acid is the most abundant hydroxycinnamic acid in plant cell wall, it is mainly linked to hemicellu⁃losic carbohydrate moieties and lignin components. Due to the unsaturation of the aliphatic side chain and its bulk, feru⁃lic acid can form stable phenoxy motieties and serve as good antioxidants. Ferulic acid is also a precursor to vanillin and other high⁃valued aromatic compounds synthesis. Ferulic acid has large number of applications in health, food, pharma⁃ceutical, and cosmetics industries. De⁃strached wheat bran contains 5 mg/g to 10 mg/g of ferulic acid and is considered as a potential resource for ferulic acid production. In this study, feruloyl esterase ( FAE) was prepared by solid⁃state fer⁃mentation of Aspergillus niger TRIIM 3�00954 derived from 10 different strains. The results showed that the maximum en⁃zyme activity was produced under the conditions as:the ratio of wheat bran to bagasse at 2∶1, the 0�6% glucose as sup⁃plementary carbon source, the 1% urea as supplementary nitrogen source, the initial pH of 6�0, and culture temperature at 30 ℃. After 7 d of fermentation, 8�2 U/g of feruloyl esterase was obtained. The optimum pH value and temperature of the FAE were 5�0 and 50℃, respectively. The optimum enzymatic hydrolysis condition is obtained as follows:tempera⁃ture 50 ℃ and pH 6�5. Under this condition, the maximum of 60�6% total ferulic acid was released from de⁃starched wheat bran after 6 h enzymatic hydrolysis using 0�6 U/g and feruloyl esterase and 90 U/g xylanase. The economic and eco⁃friendly ferulic acid recovery of phenolic acids from the agricultural and forestry by⁃products to produce value⁃added products has become a research focus in recent years. This paper provides an effective technological approach to extract ferulic acid from de⁃starched wheat bran.
