Abstract
Objective To explore the protective effect of intravenous hydrogen saturated saline on sodium taurocholate induced rat severe acute pancreatitis associated lung injury (APALI). Methods (1)The establishment and treatment of severe APALI model:54 healthy male SD rats were randomly divided into the sham operation group (Sham group),model group[severe a-cute pancreatitis(SAP)+normal saline(NS) group ] and hydrogen water treatment group(SAP+H2 group),18 cases in each group;each group was subdivided into 3 subgroups according to the time points of 6 ,12,24 h,6 cases in each subgroup. In the Sham group,the pancreas was stirred for several times after laparotomy and then the abdominal cavity was closed without conducting oth-er treatment. In the SAP+NS group and SAP+H2 group,5%sodium taurocholate(1 mL/kg) was retrogradely injected by the biliary-pancreatic duct opening (0.1 mL/min) for constructing the model,5 mL/kg normal saline or the equivalent hydrogen saturated saline was respectively injected by the tail vein at 1 h after successfully constructing the model. (2)Rats in each group were killed for collecting serum at the time points of 6,12,24 h. The serum TNF-αand IL-1βlevels were detected by ELISA. Results (1) The TNF-αlevels at 6,12,24 h were (215.3±6.0),(263.2±7.4),(288.0±5.6)ρg/mL in the SAP+NS group and (204.1±8.5), (122.4±10.3),(84.1±8.6)ρg/mL in the SAP+H2 group,which were higher than (58.8±7.5),(57.5±5.1),(59.6±6.0)ρg/mL in the Sham group,moreover the TNF-αlevel at 3 time points in the SAP+NS were higher than those in the SAP+H2 group,the differences were statistically significant(P<0.05). (2)The IL-1βlevels at 6,12,24 h were(152.8±9.8),(215.4±10.4),(254.3±8.6)ρg/mL in the SAP+NS group and (154.7±6.8),(80.3±8.3),(59.3±8.2)ρg/mL in the SAP+H2 group,which were higher than(39.8±3.3),(39.5± 3.3),(40.5±5.2)ρg/mL in the Sham group,moreover which at 12,24 h in SAP+NS group were higher than those in the SAP+H2 group,the differences were statistically significant(P<0.05),while the IL-1β level at 6 h had no statistical difference between the SAP+H2 group and SAP+NS group(P>0.05). Conclusion The tail vein injection of hydrogen saturated saline has certain treatment and protective effect on APALI.关键词
