恩度联合奥沙利铂对裸鼠胃癌种植瘤的抑制作用研究
Study on endostar combined with oxaliplatin in inhibiting transplanted gastric tumors in naked mice

Abstract
Objective:To explore the effect of endostar combined with oxaliplatin in inhibiting transplanted gastric tumors in na-ked mice.Methods:Human gastric cancer cell SGC-7901 was injected under subcutaneous armpits of naked mice until tumors appeared under their armpits.A total of 32 naked mices models were successfully established and randomly divided into blank group,oxaliplatin group,endostar group and combined group of endostar plus oxaliplatin,8 cases for each group.All group were intraperitoneally injected with drugs for continuously 14 d.During drug administration,the mice were weighed once every 72 h.Vernier caliper was used to meas-ure the maximum and minimum diameters of the lumps to calculate the relative sizes of tumors.The naked mice were sacrificed after drug administration was finished to collect the tumor tissues.Then,the tumor weight was measured to calculate tumor inhibiting rate.Im-munohistochemical staining was conducted to tumor tissues and suspicious lymph nodes,and the expression of vascular endothelial growth factor (VEGF-C)was detected.Podoplanin protein was used as lymphatic duct marker so as to calculate the positive rate of lymph node metastasis and lymphatic microvessel density (LVD).Results:Both endostar and oxaliplatin could inhibit the development of tumors,but their combination was more superior than single use in clinical efficacy.Endostar group was markedly lower than blank group and oxaliplatin group in positive rate of lymph node metastasis,count of metastatic lymph nodes and LVD,and there were signifi-cant differences (P <0.01 ),but the differences between oxaliplatin group and blank group were not significant (P >0.05).The optical density of endostar group and oxaliplatin +endostar group was significantly lower than that in oxaliplatin group and blank group (P <0.01 ).Conclusion:Endostar combined with oxaliplatin can significantly inhibit the growth of subcutaneous transplanted tumors and the metastasis of lymph nodes in naked mice,with synergic anti-tumor effect obtained.关键词
