摘要

Abstract

To increase the population density of Bacillus subtilis CN181 in the broth, fermentation media and fermentation conditions of B. subtilis CN181 were screened and optimized by single factor and orthogonal tests in this study. The results from single factor experiment showed that the optimum carbon, nitrogen sources and metal ions were corn flour, soybean dregs, K+and Na+. The best medium was composed of corn flour at 15.0 g·L-1, soybean dregs at 10.0 g·L-1, K2HPO4 at 2.0 g·L-1, and NaCl at 2.0 g·L-1 based on the orthogonal test. Finally, the culture conditions were optimized as the following:the initial pH of the fermentation medium at 8.0, the liquid medium volume at 30 mL in a 250 mL flask, the inoculation volume at 2%(v/v), the fermentation temperature at 30℃, the rotation speed at 180 r·min-1 and the fermentation time for 24 h.
