Molecular Marker Development and Bioinformatics Analysis of EST -SSR in Ryegrass

Abstract
To accelerate the application of molecular markers in ryegrass (Lolium perenne L.),the EST -SSR information analysis,molecular marker exploitation and EST function analysis were studied using ryegrass EST data (GenBank /dbEST).Among the 25 752 ryegrass EST sequences,a total of 346 microsatel-lite sequences were found,which amounted for 1.344%.The trinucleotide motifs were the most abundant SSRs (39.60%),followed by the dinucleotide motifs (31.79%).Among the trinucleotide motifs,GGC /CCG had the highest frequency of 8.38%,followed by CGC /GCG (7.51%),GCC /CGG (4.05%)and ATG/TAC (2.02%).Among the dinucleotide motifs,CT/GA had the highest frequency of 14.45%,fol-lowed by GA /CT (10.98%).According to these 346 EST sequences containing SSR,193 EST -SSR primer pairs were designed using Primer Premier 5.0 software.In which,113 (32.66%)primer pairs were found with the score values over 90.The function analysis was performed for the 113 ESTs by the BlastN and BlastX programs in GenBank.Then 72 sequences had homology with 51 kinds of proteins.关键词
