BraSDG2 RNA 干扰载体的构建和拟南芥的遗传转化

Abstract
Polyploidization is not only a natural phenomenon in plant evolution,but also an important source of plant evolution.The bioinformatics analysis has showed that BraSDG2a (Bra037086)and BraSDG2b (Bra039562)are the homologous genes of AtSDG2.Through the sequence alignment,258 bp DNA conserved fragment of BraSDG2a and BraSDG2b was used to construct the RNAi vector pFGC5941 -BraSDG2 -RNAi. Then the wild type Arabidopsis thaliana (Col -0)was transformed by floral dip method.One transgenic plant was got by PPT screening and PCR detection.The phenotypic observation on T4 generation of transgenic Ara-bidopsis showed that its biological phenotype was similar to Arabidopsis sdg2 mutant.Its plant was dwarf with few seeds.The results laid the foundation for further study on the biological function of BraSDG2.关键词
