Abstract
Objective To investigate the expression of circadian clock gene Timeless in hepatitis B virus ( HBV)-relat-ed hepatocellular carcinoma (HCC) and its correlation with hepatitis B virus x protein (HBx).Methods Immunohisto-chemistry was employed to detect the expression of Timeless in 60 cases of HCC tissues and adjacent normal tissues.Then we analyzed the relationship between the expression of Timeless and clinic pathological features in HCC patients.Meanwhile, both pcDNA3.1-HBx and pcDNA3.1 were transfected into LO2 and HepG2 cell lines, respectively.Real-time PCR and West-ern blotting were used to detect the expression of Timeless mRNA and protein.Results The protein expression levels of Timeless in HCC tissues and adjacent normal tissues were 60.0%(36/60) and 38.3%(23/60), respectively;and signifi-cant differences were found (P<0.05).The expression of Timeless in HCC tissues was associated with vascular invasion (P<0.05), was not related with sex, age, level of serum AFP (all P>0.05).HepG2 and LO2 cell lines transfected with pcD-NA3.1-HBx plasmids demonstrated higher protein and mRNA expression levels of Timeless as compared with those transfected with pcDNA3.1 plasmids (all P<0.05).Conclusion High expression of Timeless may be related to the occurrence and progression of HBV-related HCC, and HBx may be one of the reasons that causes the high expression of Timeless.关键词
