Abstract

A petroleum degrading bacterial strain SYBS01 was isolated from oil-contaminated soil,and was identified as Pseudomonas sp.Oil degradation conditions were compared with single-factor and orthogonal experimental design,with the results showing that the optimum conditions were 35 ℃,170 r · min-1,yeast extract 15 g · L-1,KH2PO4 0.5 g · L-1,and a natural pH value.Among these,the yeast extract had the greatest influence on the oil-degradation rate.The oil-degradation rate of SYBS01 increased to 85.6% four days after yeast extract had been added to the system,and was 6.7,7.8,and 2.6 times higher than were (NH4)2SO4,NH4NO3,and CON2H4,respectively.It was shown that yeast extract not only acted as a source of nitrogen,but also provided extra nutrition.A first-order kinetic model could be fitted to the oil degradation for the entire concentration range in optimum conditions.Furthermore,the half-life was found to be 26 h for oil 3 to 4 g · L-1.
