RhD mRNA剪接体全长外显子序列扩增技术的应用
Application of exon sequence amplification technique in RhD mRNA splicing

Abstract
Objective To explore the effectiveness and practicability of the RhD mRNA splicing polymorphism detection technique.Methods Two pairs of primers were designed to amplify the full-length RhD exon mRNA,to completely avoid the interference of the RHCE gene and to specifically amplify and analyze random unrelated RhD mRNA splicing in 50 cases.Results The full-length exon of RhD mRNA gene were amplified by two primers and the polymorphism of RHD gene was analyzed.The specificity of the 4 splicing-variants were amplified:exon1-3 amplification products:415bp.Exon-3-10 amplification products were composed of three different combinations:the first product was about 880bp;the second product about 750bp and the third product about 550bp.Fifty samples were analyzed:the exon-1-3 amplification products did not display deletion;the exon-3-10 amplification products with band 1 were intact;27 cases displayed exon-7 deficiency in band 2;6 cases displayed deficiency in exon-7,8;12 cases displayed deficiency in exon-8,9 in band 2;5 cases were deficient in exon-7,9.Band 3 showed complete deficiency in exon-7,8,9.Conclusion The establishment of an RhD mRNA splicing polymorphism detection technology can accurately amplify RhD mRNA gene and more effectively analyze the polymorphism of the spliced gene.关键词
