Abstract

Objective To explore the effectiveness and practicability of the RhD mRNA splicing polymorphism detection technique.Methods Two pairs of primers were designed to amplify the full-length RhD exon mRNA,to completely avoid the interference of the RHCE gene and to specifically amplify and analyze random unrelated RhD mRNA splicing in 50 cases.Results The full-length exon of RhD mRNA gene were amplified by two primers and the polymorphism of RHD gene was analyzed.The specificity of the 4 splicing-variants were amplified:exon1-3 amplification products:415bp.Exon-3-10 amplification products were composed of three different combinations:the first product was about 880bp;the second product about 750bp and the third product about 550bp.Fifty samples were analyzed:the exon-1-3 amplification products did not display deletion;the exon-3-10 amplification products with band 1 were intact;27 cases displayed exon-7 deficiency in band 2;6 cases displayed deficiency in exon-7,8;12 cases displayed deficiency in exon-8,9 in band 2;5 cases were deficient in exon-7,9.Band 3 showed complete deficiency in exon-7,8,9.Conclusion The establishment of an RhD mRNA splicing polymorphism detection technology can accurately amplify RhD mRNA gene and more effectively analyze the polymorphism of the spliced gene.
