摘要

Abstract

ObjectiveTostudytheinhibitoryeffectofEthanolExtractofHedyotis diffusaWilld.(EEHDW)onthegrowthofcolorectal cancer drug-resistant xenograft in nude mice. Methods A drug-resistant subcutaneous xenograft model was established in nude mice. Mice were randomized into 4 groups, the Control, EEHDW, 5-FU, and EEHDW plus 5-FU combination groups. The inhibition rate was calculated by the determination of tumor volume. And the expressions of PCNA, Cyclin D1, CDK4 as well as p21 were detected by RT-PCR and IHC. Results Compared with the control group, 5-FU had little effect on the tumor growth (P>0.05), while EEHDW and combination treatment significantly inhibited the growth of xenograft (P<0.05), and combination treatment had more obvious inhibitory effect than that of EEHDW. EEHDW and combination treatment significantly inhibited the proliferation of tumor cells, down-regulated Cyclin D1 and CDK4, and promoted the mRNA and protein expression levels of p21 (P<0.05). Conclusion EEHDW could inhibit the growth of colorectal cancer drug-resistant xenograft. The mechanism might be related to its down-regulation of Cyclin D1 and CDK4 and up-regulation of p21 expressions.
