甲状腺素和多奈哌齐对甲状腺功能减退症大鼠前额叶synaptotagmin-1表达的影响
Influence of thyroxine and donepezil for synaptotagmin-1 on the prefrontal cortex of hypothyroidism rats

Abstract
Objective To investigate the synaptotagmin-1 expression in the prefrontal cortex of hypothyroidism rats model and the effects of thyroxine(T4) and donepezil(DON). Methods A hypothyroidism rat model was induced by drinking 0. 05% propylthiouracil( PTU) water. All rats were randomly divided into five groups:normal control group(CON), hypothyroidism group(Hypo), T4 treatment group (T4),DON treatment group(DON),T4 com-bined DON treatment group( T4+DON) . The thyroid hormone level was evaluated by radioimmunoassay, and the expression and immune reaction product distribution of syt-1 in the prefrontal cortex of rats were analyzed respec-tively by Western blot and hypersensitivity immunohistochemical method. Results Compared to the controls, the serum thyroidhormone levels of Hypo and DON treatment group rats were significantly decreased(P<0. 05), while returned to normal in T4 and combined treatment group rats. Western blot analysis showed that the syt-1 expression in the prefrontal cortex of Hypo group, DON and T4 treatment group rats were higher than in CON group ( P <0. 05), but reached the normal level in combined treatment group. Immunohistochemical results showed that syt-1 immune reaction product was tanparticles,which was widely distributed in the prefrontal cortex of rats molecular lay-er, granular layer, lateral pyramidal cell layer, internal granular layer, the pyramidal cell layer by optical micro-scope. Results suggested that Hypo group,DON and T4 treatment group rats had significant increments of syt-1 im-mune reaction product in each layer compared with normal control group(P<0. 05), however, restored to normal levels in each layer of combination therapy group. Conclusion These findings indicate syt-1 expression in the pre-frontal cortex is increased in hypothyroidism rat model. T4 replacement therapy fails to make damage protein fully recover. Co-administration of T4 and DON leads to normalization of damage protein, which suggests that combina-tion therapy is more advantageous to hypothyroidism brain injury protein recovery.关键词
