摘要

Abstract

(S)-4-((3-chloro-4-methoxybenzyl)amino)-2-(2-(hydroxymethyl)pyrrolidin-1-yl)pyrimidine-5-carboxylate (3) is a key intermediate for Avanafil synthesis. Na2WO4 was used as a catalyst in this study and ethyl 4-((3-chloro-4-methoxybenzyl)amino)-2-(methylthio)pyrimidine-5-carboxylate (1) was oxidized by H2O2 to produce sulfone (2a) or sulfoxide (2b) derivatives. These derivatives were reacted with L-prolinol to prepare (S)-4-((3-chloro-4-methoxybenzyl)amino)-2-(2-(hydroxymethyl)pyrrolidin-1-yl)pyrimidine-5-carboxylate (3). Factors including oxidant dosage, catalyst loading, solvent and temperature on the reaction were optimized and the optimal conditions were obtained using response surface methodology, which were proved via verification experiments. The products obtained were characterized by 1H-NMR, 13C-NMR, ESI-MS and FT-IR.
