Abstract
Objective To observe the single nucleotide polymorphisms (SNPs)of +45T/G and +267G/T loci in adiponectin gene of patients with knee osteoarthritis (OA).Methods The genotypes and allele frequencies of SNPs of +45T/G and +267G/T loci in adiponectin gene from plasma of 255 knee OA patients (observation group)and 201 healthy volunteers (control group)were detected with polymerase chain reaction-based restriction fragment-length polymorphism (PCR-RFLP).Results Two SNPs met Hardy-Weinberg Equilibrium (HWE).T and G allele frequencies of +45T/G and +267G/T loci of adiponectin gene were respectively 64.6%,35.4%,and 29.2%,70.8%in the observation group, and no significant difference was found as compared with that of the control group (all P>0.05 ).But there was statistic difference between low-grade group and moderate to severe-grade group of OA patients at SNP +267G/T locus (32.9%, 67.1% and 24.1%,75.9%)(all P<0.05).Conclusion At the +267G/T locus of adiponectin gene of moderate to severe-grade OA patients,there are more GG genotype or carrying G allele.关键词
