摘要

Abstract

Objective:To establish experimental autoimmune cerebral spinal cord inflammation (EAE) model rats,and observe the pathological changes and effect of korean red ginseng extract on EAE model.Methods:30 SD rats were randomly divided into control group ,EAE ( model group ) group and KRG group.The EAR symptom score and body weight were used to evaluate the rats after the model.The inflammation of the brain and spinal cord demyelinating changes were observed by HE staining .The frequency of CD4+, CD4+/IFN-γ+,CD4+/IL-17+and CD4+/Foxp3+T cells were measured by flow cytometry ,and the expression of Foxp3,IFN-γ,IL-17 and IL-23 mRNA were evaluated by Real time-PCR in spinal cord and lymph node.Results:KRGE significantly attenuated symptom score and loss of body weight.Compared with model group , neurological impairment and pathological change were alleviated in KRG group.The number of CD4+,CD4+/IFN-γ+,CD4+/IL-17+were down-regulated in KRG,and the number of CD4+/Foxp3+was up-regulated in the spinal cord and lymph node compared with the model group ( P<0.05 ).The levels of Foxp3 mRNA was also significantly increased,while the levels of IFN-γ,IL-17,IL-23 mRNA were markedly decreased (P<0.05).Conclusion: KRG could have an effect on prevention on EAE in SD rats ,may be related to its ability to the regulations the expression levels of CD 4+,CD4+/IFN-γ+,CD4+/IL-17+and CD4+/Foxp3+.
