The Influence of Astragalus on Apoptosis Related Factors Bcl-2’s Famlily in Eld Patients with Acute Myeloid Leukemia

摘要

Abstract

Objective In order to disscuss the underlying treatment mechanism of astragalus on eld patients with acute myeloid leukemia,this study was purposed to investigate the efficacy of astragalus in the treatment of eldly patients with acute myeloid leukemia and the influence of astragalus on atopsoptosis(apoptosis related factors) Bcl-2’s famlily in the patients.Method There were 42 eld patients with AML divided into the treatment and the control groups randomly.Both of the groups were treated with chemotherapy and the treatment group received astragalus more.The rates of complete remission were analysed ,the apoptosis of leukemia cells was quantitatively examined by FCM,the expressions of Bcl-2、BCL-xl、Bax、Bak mRNA were detected by Realtime-PCR before and after therapy,The effectiveness of the two groups would be observed after treatment.Result The complete remission rate of the treatment group was statistically higher than the control one (P<0.05).The apoptosis rates of leukemia cells in the treat-ment group after the therapy were significantly higher than the control group.The mRNA expressions of Bcl-2、BCL-xl、Bax、Bak were especially different from the expressions before therapy in the treatment group (P<0.05).The expressions of Bcl-2 and Bcl-xl were notably lower than before therapy in the treatment group (P<0.05).The Bax and the Bak mRNA expressions were especially higher than before therapy in the treatment group (P<0.05).The mRNA expressions of Bcl-2、BCL-xl、Bax、Bak had no significant difference from the expressions before therapy in the control group (P<0.05).Conclusion Astragalus could promote the process of apoptosis for the leukemia cell and improve the patient’s treatment effect .The mechanism of these may be related to the increases of the apotosis for leukemia cells by reducing the expressions of Bcl-2 and Bcl-xl and promoting the expressions of Bax、Bak.
