摘要

Abstract

Melampsora larici-populina strongly endangers a variety of poplars.In order to investigate the molecular mechanisms of transcriptional regulation factors ERF3 in the reaction of the poplar resisting M.larici-populina,ERF3 genes from Populus deltoides × P.trichocarpa (less susceptible to E4 M.larici-populina) and Populus × euramericana (very susceptible to E4.larici-populina) were cloned and named as Pdt-ERF3 and Pnd-ERF3,respectively.The expression quantity change of Pdt-ERF3 and Pnd-ERF3 of before and after vaccination E4 to strong pathogenic M.larici-populina.were analyzed by employing Q-PCR technique.The expression quantity of Pdt-ERF3 reached the peak at 12-hour post inoculation (hpi).The expression quantity of Pnd-ERF3 increased significantly at 24hpi and reached the peak at 96hpi.Compared to Pdt-ERF3,the peak expression quantity of Pnd-ERF3 was delayed 84 hours.Both expression quantities of the Pnd-ERF3 and Pdt-ERF3 appeared a significant decreasing except at 168hpi,and the expression quantities of Pnd-ERF3 and Pdt-ERF3 response to rust stress were different in multiple stages.It was predicted that the expression quantity of Pdt-ERF3 was subject to the regulation of MAPK6 according to homeotic gene of Arabidopsis thaliana.This study provided a theoretical foundation for further studying ERF gene and the possible use of these genes for the production of rust resistance poplar.
