不同剂量活血、破血中药对动脉粥样硬化小鼠主动脉PTGS2、PADI4、ITGAM基因表达的影响
Effects of different doses of blood circulation and traditional Chinese medicine on the expression of PTGS2, PADI4 and ITGAM in aorta of atherosclerotic mice

Abstract
Objective To observe the effect of different doses of Chinese herbal medicine (Chinese angelica, Chi-nese medicine (Chuan Xiong), broken blood Sanleng and Ezhu) on atherosclerosis (AS) in mouse aorta cyclooxygenase 2 (PTGS2), peptidyiarginine deiminase (PADI4) and ITGAM gene expression.Methods Sixty four ApoE gene deficient mice atherosclerosis (AS) models of five weeks were randomly divided into 6 groups,the model group, atorvastatin group (the statin group), low dose of blood activating group (low blood activating group), low dose of blood breaking group (low blood breaking group), high dose of blood activating group (high blood activating group), high dose of blood breaking group (high blood breaking group).After successful modeling, drug gastric perfusion for 8 weeks, the model group was filled with distilled water, then, the mice were killed.PTGS2, PADI4 and ITGAM gene mRNA expression of the aortic tissue were tested with RT PCR method.Results Compared with model group, the PTGS2, PADI4 and ITGAM gene expression levels ( P <0.01) were significantly reduced in each drug group.(1) in the down-regulation of PTGS2 gene expression level, blood group than blood group ( P <0.01), the high dose group than in low dose group ( P <0.01), atorvastatin group was better than that of blood group and blood breaking group ( P <0.01); (2) in the down-regulation of PADI4 gene expression level, blood break-ing group is better than the live blood group ( P <0.01), the high dose group than in low dose group ( P <0.01), atorvas-tatin group was better than that of blood group and blood breaking group ( P <0.01); (3) in the down-regulation of ITGAM gene expression level, high blood group is better than the high broken blood group ( P <0.01).Low blood group and low blood group when ( P >0.05), the high dose group than in low dose group ( P <0.01), atorvastatin group was better than that of blood group and blood breaking group ( P <0.01).Conclusion Promoting blood circulation to traditional Chinese medicine and traditional Chinese medicine can reduce the expression level of PTGS2, PADI4 and ITGAM in aorta, so as to play the role of anti atherosclerosis.关键词
