益气养阴活血法对糖尿病大鼠肝脏P450的影响及其与血糖血脂的关系
Influence of Tonifying Qi,nourishing Yin and activating blood method on the cytochrome P450 in the liver and its relationship with glucose and lipid

Abstract
[Absract] Objective To explore the influence of Tonifying Qi,nourishing Yin and activating blood method on the cytochrome P450 in the liver of diabetic rats and its relationship with blood glucose and blood lipid.Methods Fifty-five rats were randomly divided into 4 groups:normal control group,model group,high-and low-dose Danzhi Jiangtang Capsule groups,and pioglitazone group.Except the control group,all others groups were fed with high-fat diet for 4 weeks,then gave diabetic model rats intraperitoneal injection with streptozotocin(STZ)of 35 mg/kg once only.The rats of high-dose DJC and low-dose DJC group were treated with DJCof 1.08 g·kg -1 ·d -1 ,0.54 g·kg -1 ·d -1 respective-ly,pioglitazone group was treated with pioglitazone of 10 mg·kg -1 ·d -1 ,the normal control group and model group were given saline 5 ml·kg -1 ·d -1 .After modeling,continuously administrate drug by drenching for 8 weeks,then put them to death and get the liver to do the tests of p450 and β-actin by the Real-Time PCR.Meanwhile,we measured the glucose,lipid and HbA1 c.Results Compared with the normal group,the expression of p450 in the model group in-creased significantly(P <0.01 ),the overexpression of p450 was significantly inhibited by DJC.compared with model group,the difference has statistical significance(P <0.01).After the model established,the level of glucose,lipid and HbA1 c were significantly increased(P <0.01),while the level of glucose,lipid and HbA1 c of high-dose DJC and low-dose DJC group were dacreased significantly.The effect of high-dose DJC and low-dose DJC were similar to pioglitazone group(P <0.01 or P <0.05).The correlation analysis showed that there was a significant positive correlation among P450 and blood glucose,HbA1 c.There was no significant correlation between P450 and lipid(P <0.01).Conclusion Danzhi Jiangtang Capsule can inhibit the activity of p450 enzyme in the liver of diabetic rats and reduce the blood glu-cose and lipid.Meanwhile,the P450 of liver and glucose is correlated.关键词
