摘要

Abstract

To filter plants with high corilagin content,a high performance liquid chromatography (HPLC)method was established for determination of corilagin contents in different plants,and the anti-gastric cancer activity of corilagin was also investigated by thi-azolyl blue (MTT)assay,acridine orange/ethidium bromide (AO/EB)staining,and Hoechst 33258 staining.HPLC analyses showed that longan (Dimocarpus longan Lour.)seed,Phyllanthus urinaria L.,longan shell,Terminalia catappa L.leaf and P .tenellus Roxb.contained higher levels of corilagin,which were 643,619,569,271 and 159μg/g respectively.MTT assay results showed that gastric cancer cells SGC7901 had IC50 value of (42.46±3.31)μmol/L with corilagin treatment.AO/EB staining and Hoechst33258 staining showed that corilagin induced apoptosis of gastric cancer cells SGC7901.As a conclusion,the conditions of HPLC explored in this study can quickly and accurately detect corilagin content of different plants,and longan seed contains the highest level of corila-gin among the 1 2 kinds of plant samples,and corilagin exhibits anti-gastric cancer activity.
