摘要
Abstract
Objective Observation the effect of arsenic trioxide (ATO)on the growth of human fibroblast-like synoviocytes (FLSs),and to explore the potential therapeutic effects of ATO on rheumatoid arthritis (RA).Methods Cultured primary human joint FLSs and passaged 3-5 generations,treated cells with different concentration of ATO,detected TNF-ɑ、IL-1 in supernatant with ELISA,and detected cell growth status in different treatment groups by MTT.Results After treated with different concentration of ATO,ELISA results showed that TNF-ɑ,IL-1 levels in test groups were significantly lower than those of the control group (P<0.05).MTT results showed that with the drug concentration increased and treatment time prolonged,the enhanced the inhibitory effect of ATO on FLSs.Conclusion ATO can inhibit the growth of human FLSs,inhibit the secretion of synovial cells,and then produce intervention on RA inflammation and immune process,suggesting that it can be a potential drugs for RA treatment.关键词
