大鼠脑缺血再灌注后海马组织中GFAP和p-STAT3表达研究
Expression of GFAP and p-STAT3 in rat hippocampus following focal cerebral ischemia-reperfusion injury

Abstract
Objective To detect the proliferation of astrocytes and the expression of phosphorylated STAT 3(p-STAT3) in the dentate gyrus ( DG ) of rat hippocampus after cerebral ischemia-reperfusion ( I/R ) , and to provide reference for regulation of the abnormal activation of astrocytes .Methods A focal cerebral I/R model was induced by an intraluminal filament embolism.Forty-eight SD rats were randomly divided into the sham-operated (Sham) group, the middle cerebral artery occlusion-reperfusion 24 h (I/R-24 h) group, 72 h (I/R-72 h) group and 7 d (I/R-7 d) group.The expression of GFAP in positive cells and p-STAT3 at each time point after I/R was detected by Western blotting and immunofluorescence .Results Compared with the Sham group , the expression of GFAP in the hippocampus increased in both I/R-72 h group and I/R-7 d group (P<0.05).GFAP expression was upregulated gradually after I/R and there were statistically significant differences between three time points (24 h, 72 h and 7 d)(P<0.05).Compared to Sham group, the level of p-STAT3 expression of the hippocampus at various time points after cerebral I /R was higher (P<0.05).The p-STAT3 expression was the highest at 24 h, and then decreased at 72 h.There was still a certain amount of p-STAT3 protein at 7 d following reperfusion.Compared with I/R-24 h group, p-STAT3 contents decreased significantly in both I/R-72 h and I/R-7 d groups, and the difference was statistically significant (P<0.05).Conclusion After cerebral I/R injury, the cell proliferation and expression of p-STAT3 in astrocytes are significantly increased in the hippocampus .The peak of p-STAT3 expression precedes astrocyte proliferation .关键词
