摘要

Abstract

Foxtail millet is drought-tolerant crop,but the molecular mechanisms related to drought-tolerant are still un-clear.[Objective]This study aimed to determine whether the drought-tolerant character of foxtail millet was related to the key enzymes of starch metabolism and lay the foundation for the further study on the molecular mechanism of drought re-sistance in foxtail millet and drought resistance breeding of foxtail millet.[Methods]Bioinformatics analysis was applied to analyze the basic sequence characteristics of SBD gene family and the promoter elements in foxtail millet,built the phylo-genetic tree of the gene family and determine the evolutionary relationships with other species.Based on digital gene ex-pression profiling data,we analyzed the expression characteristics of SiSBD gene in drought-tolerant variety ‘Gou Gou Mu Ji Ju (GG)’and drought-sensitive variety ‘Jin Fen 1 6 (JF-1 6 )’under PEG stress.[Results]The genome sequence data of foxtail millet showed that there were five members of SiSBD gene family named SiSBD1 ,SiSBD2 ,SiSBD3 , SiSBD4 and SiSBD5 .The sequence analysis indicated that the length of encoded protein sequence was in the range of 3 60~949 amino acid residues,the molecular weight was 3 9 ~ 1 08 KD,the isoelectric point was 4.1 8~6.72. The Cis-act-ing elements associated with drought tolerance were identified including Cis-acting elements related to MYB gene regula-tion,plant hormones as MEJA,ETH,SA,ABA,and plant defense and abiotic stress response regulation.The phyloge-netic tree showed that SiSBD1 and SBD in Sorghum bicolor and Zea mays were classified to the same group;SiSBD2 and SBD2 ,SBD3 in Zea mays were classified to the same group;SiSBD3 ,SiSBD4 and SiSBD5 were classified to the same group.The analysis results of expression of starch binding domain gene family in foxtail millet under PEG stress showed that the expression of SiSBD1 increased in drought-tolerant variety ‘Gou Gou Mu Ji Ju’and decreased in drought-sensitive variety ‘Jin Fen 1 6’,the expression of other four genes decreased in both varieties.[Conclusion]These results suggested that the SDB gene families related with response to drought stress and laid a theoretical foundation for breeding drought tolerant variety in the future.
