Developed specific-markers and sequence analysis related with specific loci among 40 cultivars of Buck-wheat by SRAP-PCR molecular marker system

摘要

Abstract

[Objective]Most of these Buckwheat cultivars,its seeds were heterozygosity because of their selfing incom-patibility.Hence,it has important product value and reality significance that developed molecular marker to identify different varieties of Buckwheat cultivars and seeds purity.[Methods]According to these DNA polymorphism loci a-mong 40 Buckwheat cultivars through SRAP-PCR system,we recycled and resequenced these different DNA loci,and analyzed their sequence character by bioinformation method.[Results]8 DNA polymorphism loci were cloned.Only 2 loci could find forward and reverse primer sequences and single primer in any other loci.So the 2 loci sequence were named 8801-1 and Mhua 5. The sequence length of 8801-1 was 705bp,and included 3 restriction Enzyme cutting site. 1 ORF domain was predicted in this sequence.Through BLASTP analysis,it had not find any homologous protein with this sequence.The sequence length of Mhua5 was 687bp,and included 3 restriction Enzyme cutting site.2 ORF do-main was predicted in this sequence.Through BLASTP analysis,there was 71% homology between the ORF2 in buck-wheat and retrotransposon pol protein in Arabidopsis thaliana .[Conclusion]The results suggested that this sequence could be included a retrotransposon element of gene.In summary,we have got 2 special DNA loci related with 2 Buck-wheat cultivars.One locus included retrotransposon element,anther locus was redundancy genome sequence.We hope to develop the 2 loci for STS markers,and apply them to analyze genetic diversity,and identify seeds purity,and con-struct genetic mapping and marker-assisted selection (MAS)of Buckwheat cultivars.
