响应曲面法优化石榴皮总黄酮提取工艺
Optimized Extraction Process of Total Flavonoids from Pomegranate Peel by Response Surface Methodology

Abstract
This paper investigated the optimum technological conditions for extracting total flavonoids from pome-granate peel by ultrasound-assisted extraction process.Key factors were selected by Plackett-Burman design, which could influence total flavonoids extraction efficiency from pomegranate peel .Steepest ascent experiment was conducted to determine the optimal level of these factors.The response surfaces were delineated based on the central composite design model in Design-Expert 8.0.6 software.The results showed that the initial temperature and ex-traction time had no significant effect on the extraction of total flavonoids from pomegranate peel within the scope of the research institute.Extraction at room temperature for 10 min,the optimum extraction conditions were deter-mined to maximize the extraction of total flavonoids from pomegranate peel:the liquid-to-solid ratio (mL/g),the proportion of ethanol and the ultrasonic power was 82:1,65% and 154 W,respectively.Under this condition,5 par-allel verification tests were carried out and the average value of total flavonoids extraction (165.09 mg/g)reached 96. 42% of the theoretical prediction value (171.22 mg/g).It showed that the process was reliable with higher extrac-tion rate,so it could be used as a reference to extract the total flavonoids from pomegranate pee on large scale.关键词
