Gen mutations of Epstein-Barr virus-encoded latent membrane protein 1 in pediatric lymphomas of Gingdao area

Abstract
Objective To understand the gene polymorphisms of Epstein-Barr virus (EBV)-encoded latent membrane protein 1 (LMP1)gene in pediatric patients with lymphoma from Qingdao area and to investigate the significance of LMP1 in the development of pediatric lymphomas.Methods Nested-PCR and DNA sequencing were used to analyze the gene se-quences of LMP1 in isolates from 66 cases of EBV-positive samples of children with lymphomas.The sequences of LMP1 were classified according to the signature changes and the phylogenetic tree.The distribution of LMP1 variants in pediatric lymphoma cases was compared with the previous data from adult NK/T cell lymphoma cases and healthy donors in the same area.Results Three subtypes of LMP1 gene,China 1,China 2 and Med-,were observed in 66 isolates,with distribution of 84.8%,7.6% and 6.1%,respectively.Another 1 (1.5%)isolate was recombinant strain of China 1 and China 2. The frequencies of XhoI(-)variant (loss XhoI site in N-terminus of LMP1)and prototype XhoI(+)were 93.9% and 6.1%,respectively,and those of del-LMP1 variant (30 bp deletion in C-terminus of LMP1)and prototype wt-LMP1 were 84.8% and 15.2%,respectively.The distribution of LMP1 variants in pediatric lymphoma was similar to that in adult NK/T cell lymphoma,while the frequency of China 1,XhoI(-)or del-LMP1 in children with lymphoma was significantly higher than that in healthy donors (all P <0.05).Conclusion China 1,XhoI(-)and del-LMP1 are the major LMP1 subtypes in pediatric lymphomas from Qingdao and may associate with the susceptibility of pediatric lymphomas.关键词
