Changes and clinical significance of maternal plasma sTRAIL levels, placental tropho-blast TRAIL and TRAIL-receptors expression in preeclampsia

Abstract
Objective:To evaluate the changes of maternal plasma sTRAIL levels,pla-cental trophoblast TRAIL and TRAIL-receptors expression in preeclampsia and to explore their clinical significances in the pathogenesis of preeclampsia. Methods:Fifty-six preeclampsia pregnancies were enrolled as the research group,which were divided into the research group 1 (31 cases of severe preeclampsia pregnancies) and the research group 2 (25 cases of pre-eclampsia) . As normal control,30 healthy pregnant women (28 to 36 weeks of gestation) were enrolled as control group 1,30 healthy pregnant women (36+1 to 38 weeks of gestation) as the control group 2 ,27 pathologic pregnancies includes preterm labor or premature rupture of mem-branes (28 to 36 weeks of gestation) as the control group 3 and 30 pathologic pregnancies(36+1 to 38 weeks of gestation) as the control group 4. Venous blood samples and the placenta tissues were collected from both research groups. As control,we took venous blood samples from the control group 1 and 2 , placental tissues from the control group 3 and 4 . Maternal plasma sTRAIL levels were detected by ELISA. TRAIL and TRAIL-Rs ( DR4、DR5、DcR1、DcR2 ) ex-pressions in placental trophoblast were detected using immunohistochemical. Results:Compared to control groups, the levels of maternal plasma sTRAIL were significantly lower in the pre-eclampsia groups. Significant difference between the sever preeclampsia group and control group 1 (P<0. 001),between the preeclampsia group2 and control group 2(P<0. 005) were found. Maternal plasma sTRAIL level was also lower in the group1 than that in the group2 ( P<0 . 05 ) . Immunohistochemical staining of placental tissues showed higher expressions of DR4 and DR5 but lower expressions of DcR1 and DcR2 in the research groups compared to the control groups. Compared to the group 2,the expressions of DR4 and DR5 were higher in the severe preeclampsia(P<0. 05) and with no significant difference of DR1 or DR2 expressions was found. Conclusions:Maternal plasma sTRAIL levels and placental TRAIL/TRAIL-Rs expres-sions changes significantly in preeclampsia women. The imbalance of TRAIL/TRAIL-Rs may participate in the pathological process of preeclampsia.关键词
