摘要

Abstract

Objective:To investigate the effects of long-coding RNA H19 on proliferation,migration and invasion of pancreatic cancer cells.Methods:We first detected the expression of H19 in Patu8988, SW1990,Bxpc3 and Panc-1 pancreatic cancer cells with real-time quantitative PCR (qRT-PCR).The SW1990 and Bxpc3 cells had higher H19 expression while Patu8988 and Panc-1 had lower H19 expression. The H19 siRNAs were transfected into pancreatic cancer cells SW1990 in vitro.The mRNA expression of ZEB1 and mir-200 were analyzed with qRT-PCR.The effects of H19 on proliferation were evaluated by CCK-8 and colon formation assays.The expression of cell proliferation-and EMT-related proteins were de-tected with Western blotting.Results:Compared with the control group,SW1990 cells transfected with H19 siRNAs revealed a significant lower level of H19 and lower the ability of cell colony formation.Howev-er,there was no significant change on cell proliferation rate;the expressional levels of ZEB1,Snail and PC-NA were up-regulated as well as the ratio of Bcl-2 to Bax;the mRNA level of ZEB1 was up-regulated while the expression of miR-200 showed no significant change.Conclusion:Down-regulation of H19 promotes cell proliferation,migration and invasion in pancreatic cancer cells SW1990.
