人HP1α/pEGFP-N1真核表达载体的构建及其表达和定位

Abstract
Objective: To observe the expression and localization of human HP1α, an eukaryotic expression vector HP1α/EGFP-N1 was constructed and introduced into HeLa cells. Methods:HP1αcDNA were obtained by RT-PCR, and cloned into the eukaryotic expression vector pEGFP-N1 to construct HP1α/EGFP-N1 that was transfected into HeLa cells with Lipofectamine 2000. The expression and location of HP1αin transfected cells was detected by fluorescence microscope. Results: Diagnostic restriction digestion and sequencing analysis verified the identity of the plasmid, HP1α/EGFP-N1. After transfection, HP1α is localized in the nucleus of HeLa cells. Conclusion: Eukaryotic expression plasmid, HP1α/EGFP -N1 is successfully constructed and expressed effectively in HeLa cells.关键词
