Abstract
Objective:To investigate the effect of hypoxia on the proliferation and inva-sion of human trophoblast cell line HTR-8/SVneo,and explore the possible molecular functional mechanism. Methods:HTR-8/Svneo cells were divided into two groups:cultured in hypoxic conditions and normoxic conditions. CCK8 test and Transwell assay were performed to detect the proliferation and invasion ability of these two groups. Used RT-PCR and Western blot to detect the expressions of HIF-1α mRNA,p-TrkB,TrkB,p-Akt and Akt. Results:The proliferation and invasion of HTR-8/SVneo cell were significantly inhibited after treatment with hypoxia ( P<0. 05). Long duration of hypoxia treatment reduced the expression of HIF-1α mRNA,and the expression of p-TrkB was also significantly inhibited. Moreover,the phosphorylation level of Akt was significantly inhibited ( P<0 . 05 ) . Conclusion:Long duration of hypoxia treatment inhibi-ted the proliferation and invasion of HTR-8/SVneo by inhibiting the activation of TrkB,which led to inhibition of PI3K/Akt signaling pathway.关键词
