Abstract
Objective To explore the role of kisspeptin/kiss1r system on follicular developmental disorder in the ovaries of polycystic ovary syndrome (PCOS) rats .Methods The PCOS rat models were constructed by Letrozole with intraperitoneal in‐jection .Serum hormone levels and morphological changes of ovary were examined by ELISA and H‐E staining ,respectively . The expression of kisspeptin/kiss1r system and FSHR in experimental rat ovaries was detected through immunohistochemistry . Results Compared with the normal group ,body weight of the PCOS group increased significantly from the beginning of the tenth day after filling the stomach .Simultaneously ,LH/FSH ratio was considerably increased ,which lead to hyperandrogenism . Small follicles and atresia follicles were occupied the ovarian cortex of PCOS and most of the follicles showed cystic dilatation whose cell layers were significantly reduced .FSHR and kisspeptin/kiss1r were expressed in the cytoplasm of ovaries ,and kisspeptin/kiss1r was mainly expressed in granulosa cells ,membrane cells and corpora luteum ,whereas FSHR was expressed only in granulosa cells .The expression of kisspeptin/kiss1r and FSHR in the PCOS group was significantly decreased .Conclu‐sion Kisspeptin/kiss1r system expressed in the ovaries may play an important role in the process of follicular developmental disorder ,which provides a novel idea for clinical treatment and basic research of PCOS .关键词
