八宝丹处理肺腺癌细胞A549和SPCA-1对顺铂化疗敏感性的影响
Effect on sensitivity to cisplatin after Babaodan treatment of human lung adenocarcinoma cells A 549 and SPCA-1
摘要
Abstract
Objective To study the babaodan treatment of human lung adenocarcinoma cell line A 549 and line SPCA‐1 to cis‐platin (DDP) chemotherapy sensitivity .Methods Collected human lung adenocarcinoma A549 and SPCA‐1 cell lines for differ‐ent treatment ,were randomly divided to control group (or not) ,DDP group (4 UM ) ,babaodan DDP group (0.75 mg/kg+4 UM) ,treatment was 3 weeks ,used MTT assay to detect the ability of cell proliferation ,used terminal deoxynucleotidyl transfer enzyme mediated D UTP nick end labeling determination (TUNEL) method to detect apoptosis rate ,used Transwell assay to detect the ability of cell migration .Results Compared with the control group ,the A549 and SPCA‐1 cell proliferation of DDP group and babaodan+DDP group decreased significantly (P<0.01);compared with the DDP group ,the A549 and SPCA‐1 cell proliferation of babaodan+DDP group had no significant difference (P>0.05) .And compared to the control group ,the DDP group and babaodan+DDP group of A549 and SPC‐A‐1 cells apoptosis increased significantly (P<0.01);compared with the DDP group ,babaodan+DDP group on A549 and SPC‐A‐1 cells apoptosis increased significantly (P<0.05) .And compared to the control group ,A549 and SPC‐A‐1 cell migration number of DDP group and babaodan+DDP group decreased significantly (P<0.01);compared with the DDP group ,babaodan+DDP group on A549 and SPC‐A‐1 cell migration number decreased sig‐nificantly (P<0.001) .Conclusion Babaodan DDP can increase the apoptosis of cancer cells ,reduces the proliferation of cancer cells ,reduces cells and the number of migrated of the A 549 and SPC‐A‐1 ,and enhances the chemotheropy sensitivity of DDP .关键词
