摘要

Abstract

Objective To study the protective effects of pituitary adenylate cyclase activating polypeptide ( pitui-tary adenylate cyclase activating polypeptide , PACAP) and its derivative peptide on the thymus microenvironment func-tions cell injury induced by dexamethasone .Methods The primary cells were processed by dexamethasone (Dex).The proliferation and cytoxicity assay kit (CCK8) were applied for detecting the cell activity of processed cells .Annexin V-FITC/PI double staining flow cytometry was applied for detecting the cell proptosis .ELISA was applied to measure the ex-pression of cytokines in co -culture system.Results The activity of processed cells was significantly reduced .Flow cy-tometry showed significantly increase survival and reduction in apoptosis in PACAP -PTD and PACAP groups ( P <0.05).ELISA results showed that the content of cytokines in dosing group were also significantly improved .Conclusion PACAP-PTD and PACAP protect the on the function cells of thymus microenvironment from apoptosis induced by dex -amethasone , and PACAP-PTD is more apparent than PACAP .
