结肠癌中干细胞标记物的表达情况与病理特征和预后的关系研究
Study on the Relationship Between Stem Cell Marker in Colon Carcinoma Expression and Clinicopathological Characteristics and Prognosis

Abstract
objective To study the stem cell marker Lgr5(rich in leucine repeated sequences of G protein coupled receptor 5 (leucine rich repeat-containing the G protein-coupled receptor 5,Lgr5),also known as 49 G protein coupled receptor(the G protein coupled receptor49,GPR49)protein expression levels in human colorectal carcinoma tissue and its clinical pathological characteris-tics,the evolution process of colorectal cancer and the relationship between significance.Methods To collect common surgical re-section or endoscopic biopsy of 30 cases of normal colon mucosa,colonic tumor 177 cases and 210 cases of colon cancer clinical tissue specimens,all cases were diagnosed by pathological,preoperative not put,chemotherapy;preserved specimens with paraffin embedding.Using immunohistochemical method (Immunohistochemistry,IHC)blood test the expression of CD133 in tissue samples, Lgr5,and analyzes its relationship with clinical pathological indicators.Results Tumor and colon cancer was 13.33%(4/30), 42.94%(76/177)and 73.33%(154/210),expression in colorectal cancer and adenoma were significantly higher than normal colonic mucosa (χ2=42.0130,χ2=42.0130,P<0.05),but expressed in colon cancer group was higher than adenoma (χ2=36.8039,P<0.05). Lgr5 colon tissues in the control group,the colonic tumor group and the positive expression rate of colon cancer group were respec-tively 0,23.73%(42/177)and 57.14%(120/210),expression in colorectal cancer and adenoma were significantly higher than normal colonic mucosa (χ2=34.2857,χ2=34.2857,P<0.05),but expressed in colon cancer group was higher than adenoma (χ2=44.0620,P<0.05),colorectal cancer tissues Lgr5 protein expression level and patient gender,age,tumor growth,gross morphology,tissue type no correlation (P>0.05),the expression of CD133 molecules associated with patients' survival.Conclusion CD133,Lgr5 expressed in colon cancer tissue were higher,shows that the formation of colon cancer stem cells related,CD133,Lgr5 may play a role in the pro-cess of tumorigenesis,and associated with the progress of tumor,and can be used as a molecular marker of early diagnosis.关键词
