Effects of Psoralen Flavonoids on β-amyloid25-35 Neural Stem Cell Proliferation and ApoptosisΔ

Abstract
OBJECTIVE:To probe into the effects of psoralen flavonoids on β-amyloid Aβ25-35 neural stem cell proliferation and apoptosis .METHODS:The third generation of neural stem cell suspension was extracted to be divided into alzheimer disease ( AD) mode group, experimental group , antagonist group and blank control group .Aβ25-35 5μmol/L was added into AD mode group cells to establish neural stem cells model of alzheimer ’s disease; Aβ25-35 5μmol/L and psoralen flavonoids were added into experimental group cells; Aβ25-35 5 μmol/L, psoralen flavonoids combined with estrogen receptor antagonism were added into antagonist group cells ;solution was added into the blank control group cells; respectively inoculated into a 96-pore plate .Methyl thiazolyl tetrazolium ( MTT ) method was adopted to detect the effects of psoralen flavonoids on Aβ25-35 injured neural stem cell proliferation , Western blot was adopted to detect the changes of Caspase-3 activity after the intervention of psoralen flavonoids;and effects of psoralen flavonoids on Aβ25-35 injured neural stem cells apoptosis . RESULTS: According to MTT, the absorbance of experimental group cells were significantly higher than that of AD mode group in 12 h, 24 h, 48 h, 72 h and 7 d the difference was statistically significant ( P<0.05) .The expression of Caspase-3 of experimental group were significantly lower than that of AD mode group in 1 d, 2 d, 3 d, 7 d, 14 d and 21 d, the difference was statistically significant (P<0.05).CONCLUSIONS:Psoralen flavonoids can provide protect effects on Aβ25-35 injury rat hippocampus neural stem cells by anti-apoptotic mechanisms , the effects are induced by estrogen receptor .关键词
