一种通用高效液相色谱测定植物叶片内源激素水杨酸的方法
A general method for detecting the content of endogenous salicylic acid in plant leaf by HPLC

Abstract
To accurately determine salicylic acid ( SA) level in plant, methanol was used to extract SA from Arabidopsis leaf, with 3-hydroxy benzoic acid being the internal standard. After acid hydrolysis, the hydrolysate was partitioned with ethyl acetate and hexane mixture (1 : 1), and followed by high performance liquid chromatography (HPLC) analysis under different ratios and pH of the mobile phase. Results showed that SA was thoroughly separated from the complex mixture when methanol accounted for 10% of sodi-um acetate and methanol mixture under pH 6.0. Furthermore, rice and narcissus leaf was used to verify this method, whose results confirmed that this method was generally applicable for SA detection in plants.关键词
