盐酸氨基葡萄糖对膝骨关节炎膝关节软骨下骨Ⅰ型胶原及骨钙素表达的影响
Effect of glucosamine hydrochloride on type I collagen and osteocalcin level in knee subchondral bone of patients with osteoarthritis

Abstract
Aim:To research the effect of glucosamine hydrochloride on type Ⅰcollagen and osteocalcin level in knee subchondral bone of patients with knee osteoarthritis .Methods:Forty-six postmenopausal female patients with knee osteoar-thritis underwent TKA surgery were allocated into 2 groups, those accepted glucosamine hydrochloride oral administration for 12 weeks during the half year before surgery were observation group , and those without the glucosamine hydrochloride treatment were control group .The subchondral bone tissue (0.5 cm3 ) of tibial in the 2 groups was collected and detected by micro-CT, collagen picric acid sirius red staining , osteocalcin immunohistochemical staining , as well as collagen Ⅰand os-teocalcin Western blot detection .Results:Compared with control group , the relative bone volume , intertrabecular spaces , trabecular thickness and numbers of trabecular all improved in the observation group .According to collagen picric acid siri-us red staining and immunohistochemical staining , the contents of type Ⅰcollagen and osteocalcin in the observation group were higher than those in control group (P<0.05).According to the results of Western blot ,the expression levels of typeⅠcollagen and osteocalcin in the observation group were higher than those in control group (P<0.05).Conclusion:Glu-cosamine hydrochloride can effectively improve the subchondral bone microstructure and function of knee in female patients with knee osteoarthritis by regulating the expressions of type Ⅰcollagen and osteocalcin in subchondral bone tissue .关键词
