高血压小鼠心肌肥厚过程中心肌组织 Prx-2、8-OHdG 蛋白表达变化及意义
Expression changes of Prx-2 and 8-OHdG protein in mice with hypertension-induced myocardial hypertrophy

Abstract
Objective To investigate the expression and significance of peroxiredoxin-2 ( Prx-2) and 8-hydroxy-2′-de-oxyguanosine (8-OHdG) in mice with hypertension-induced myocardial hypertrophy .Methods Sixty male mice were ran-domly divided into the observation group (n=40) and control group (n=20).Mice in the observation group received 2-kidney1-clip (2K1C) to make the models of mice with hypertension-induced myocardial hypertrophy , while there was no silver clip in mice of the control group .The systolic blood pressure ( SBP) was measured in all mice before the operation , and 2, 4, 8 and 12 weeks after 2K1C.Ten mice in observation group and five mice in the control group were scarified at 2, 4, 8 and 12 weeks after surgery to measure the whole heart quality index and left ventricle quality index .Prx-2 and 8-OHdG protein expression was detected by Western blotting and Immunohistochemistry .And then, we analyzed the correla-tion between Prx-2 and 8-OHdG.Results No significant difference was found in the preoperative SBP between the control and observation groups (P>0.05).Compared with the control group , SBP, whole heart quality index and left ventricle quality index was increased in the observation group at 2, 4, 8 and 12 weeks after surgery, indicating that the hypertension model was successful (all P<0.05).The expression levels of 8-OHdG and Prx-2 protein in the observation group at 2, 4, 8 and 12 weeks after surgery were significantly higher than those of the control group (all P<0.05), and the expression levels of 8-OHdG and Prx-2 protein was positively correlated with each other ( r=0.95, 0.97, 0.97 and 0.95, P<0.05).Conclusion The expression levels of Prx-2 and 8-OHdG in mice with hypertension-induced myocardial hypertro-phy are increased , and they participate the occurrence and development of myocardial hypertrophy through oxidation and anti-oxidation.关键词
