Abstract
Objective To investigate the effects of American cockroach extract (ACE) enema on the expression of in-sulin-like growth factor-1 (IGF-1) and transforming growth factor-β (TGF-β) in the colonic mucosa of rats with ulcerative colitis (UC) induced by 2,4,6-trinitrobenzene sulphonic acid(TNBS). Methods Thirty healthy male Wistar rats were randomly divid-ed into the control group(8 cases) and model group(22 cases) according to the random number table method in September 2015. Six cases in the model group were randomly killed by the random number table method after constructing the UC model by TNBS. The colonic changes were observed under the microscope for confirming the model construction success ,other 16 cases were di-vided in to the research group and model group according to the random number table method ,8 cases in each group. The re-search group was given ACE 5 mL/(kg·d)by retention enema ,while the model group and control group were given the same vol-ume of normal saline by the retention enema,once daily for continuous 10 d. The disease activity index (DAI) was calculated after 10 d enema,colonic gross morphology and histological damage scores were performed. The expressions of IGF-1 and TGF-βwere detected by real time reverse transcription-polymerase chain reaction(RT-PCR) and immunohistochemistry. Results Compared with the model group,DAI,colonic gross morphology and histological damage scores in the research group were improved signifi-cantly(P<0.05),the expressions of IGF-1and TGF-βin the research group were significantly lower than those in the model group, the difference was statistically significant ( P<0 . 05 ) . Conclusion ACE has better curative effect in the TNBS induced UC rats,one of possible mechanisms is down-regulating the expression of IGF-1 and TGF-βin colonic mucosa.关键词
