自发性高血压大鼠外周血淋巴细胞连接蛋白43与炎性因子表达水平的相关性研究
Correlation between Connexin 43 in Peripheral Blood Lymphocytes and Expression Level of Inflammatory Factors among Spontaneously Hypertensive Rats

Abstract
Objective To analyze the relationship between Cx43 in CD +4 and CD +8 T cells in peripheral blood and levels of IL-2 and IL-6 among spontaneously hypertensive(SH)rats. Methods During April 2015 to April 2016,20 rats with SH rats and 20 Wistar Kyoto(WKY)rats with normal arterial pressure were selected as experimental animals. The peripheral blood from aorta abdominalis was collected and centrifuged,the plasma was used to detect levels of IL-2 and IL-6 by using ELISA method,the lower blood cells were used to separate peripheral blood lymphocytes. A total of 5 × 105 lymphocytes were collected, the positive expression rate of Cx43 in CD +4 and CD +8 T cells was detected by the flow cytometer. The lymphocytes were divided into blank control group,Concanavalin A(ConA)group(5 μg/ ml ConA was added after 48 hours of culture),and Gap27 +ConA group(500 μmol/ L Gap27 was added before culture,5 μg/ ml ConA was added after 48 hours of culture). RNA was extracted from lymphocytes,the expression levels of IL-2 mRNA and IL-6 mRNA were detected by real - time RT - PCR. Results The expression level of IL-2〔(152. 40 ± 29. 62)pg/ ml〕among SH rats was significantly higher than that〔(73. 41 ± 20. 16)pg/ ml〕among WKY rats(t = 7. 24,P < 0. 05),the expression level of IL-6〔(29. 74 ± 1. 90)pg/ ml〕among SH rats was significantly higher than that〔(22. 58 ± 1. 06) pg/ ml〕among WKY rats( t = 14. 72,P < 0. 05). The positive expression rate of Cx43 in CD +4 T cells〔(3. 48 ± 0. 58)% 〕among SH rats was significantly higher than that〔(1. 36 ± 0. 18)% 〕among WKY rats(t = 15. 61,P < 0. 05),the positive expression rate of Cx43 in CD +8 T cells〔(39. 02 ± 4. 77)% 〕among SH rats was significantly higher than that〔(27. 48 ± 1. 90)% 〕among WKY rats(t = 10. 05,P < 0. 05). For SH rats,there were significant differences in expression levels of IL-2 mRNA and IL-6 mRNA among different groups( P < 0. 05),the expression levels of IL-2 mRNA and IL-6 mRNA in ConA group were significantly higher than those in blank control group and Gap27 + ConA group,respectively,the expression level of IL-6 mRNA in Gap27 + ConA group was significantly lower than that in blank control group(P < 0. 05). For WKY rats,there was significant difference in expression level of IL-2 mRNA among different groups(P< 0. 05),the expression level of IL-2 mRNA in ConA group was significantly higher than that in blank control group and Gap27+ ConA group,respectively( P < 0. 05). Conclusion The level of inflammatory factors increases because of inflammatory responses among SH rats. After the information communication between lymphocytes is blocked by carbenoxolone,the levels of inflammatory factors significantly decreases,and gap junction may be involved in the inflammation reaction of hypertension.关键词
