Expression of obestatin in circulation and kidneys of rats with heart failure after myocardial infarction and its biological significance

Abstract
Objective To investigate the expression of obestatin in the circulation and kidneys of heart failure rats after myocardial infarction and its possible biological significance. Methods A heart failure rat model was induced by coronary artery ligation. Enzyme-linked immunosorbent assay was used to test the plasma concentration of obestatin. Immunohistochemistry was used to detect the expression of obestatin in the rats' kidneys and integral optical density (IOD) was measured to analyze the difference among different groups. The expression level of obestatin in the kidneys was measured by Western blot and compared among different groups. Results The plasma concentration of obestatin in the heart failure group was significantly higher than that in the normal group and the sham group ( < 0.05). Immunohistochemistry proved that obestatin was expressed in renal tubules and collecting tubules. The level of obestatin in the kidneys of the heart failure group was significantly higher than that in the normal group and the sham group ( <0.05). Conclusions The expression of obestatin increases in the circulation and kidneys of rats with heart failure. Obestatin might participate in the development of water retention in heart failure by endocrine and autocrine or paracrine secretion.关键词
